A model for floral color inheritance in Leucaena (Leguminosae) Genet. Mol. Biol.

A population of 513 hybrids of Leucaena leucocephala (4x) and L. diversifolia ssp. diversifolia (4x), that is being analyzed for several morphological and agronomical characteristics, aiming at selecting cold tolerant plants, was also analyzed for floral color. A model of additive dominant inheritance (pink color dominant over white color, or presence versus absence of pigment, and the number of dominant alleles present determining the intensity of the pink color) is proposed. The action of two independent locus sets is also suggested, one for the male (with two main structural genes) and another for the female floral organs (one main structural gene). It is suggested that an undetermined number of regulatory genes affect color expression.

Chromosome studies of some Vernonia species (Asteraceae) Genet. Mol. Biol.

Chromosomes of 12 Vernonia species (Asteraceae) from South America were studied. First counts were reported for: V. alpestris Gardn. (n = 17), V. chalybaea Mart. ex DC. (2n = 32), V. cotoneaster Less. (2n = 30), V. fruticulosa Mart. ex DC. (n = 16) and V. lithospermifolia Hieron. (n = 10, 2n = 20). Furthermore, in V. aurea Mart. ex DC. (2n = 32), V. mollissima D. Don (n = 32) and V. saltensis Hieron. (2n = 64) numbers different from those reported in the literature were found. Karyotypes were described for some species. Screening cotton genotypes for seedling drought tolerance Genet. Mol. Biol.

The objectives of this study were to adapt a screening method previously used to assess seedling drought tolerance in cereals for use in cotton (Gossypium hirsutum L.) and to identify tolerant accessions among a wide range of genotypes. Ninety genotypes were screened in seven growth chamber experiments. Fifteen-day-old seedlings were subjected to four 4-day drought cycles, and plant survival was evaluated after each cycle. Three cycles are probably the minimum required in cotton work. Significant differences (at the 0.05 level or lower) among entries were obtained in four of the seven experiments. Apomixis maintains heterosis and avoids transmission of systemic pathogens which accompany vegetative propagation of cassava. An embryonic study of cleared ovules of two cassava clones in toto showed them to be of aposporic nature. Cytogenetic analysis of the two clones revealed an aneuploid structure (2n + 1) in apomictic individuals, whereas it was 2n in the sexually reproduced plants.

Genetic variability of three Pimelodidae species, Pimelodus maculatus, Iheringichthys labrosus, and Pinirampus pirinampu, collected at one site in the Tibagi River, was comparatively analyzed using protein data for six systems which code 15 loci in liver, muscle, and heart. The proportion of polymorphic loci (<img src="http:/img/fbpe/gmb/v21n4/21n4a14.jpg" alt="21n4a14.jpg (768 bytes)" align="middle">) for P. maculatus, I. labrosus, and P. pirinampu was 13.33, 20, and 6.67%, respectively, and mean heterozigosity was 6, 8.3, and 4.3%. Purine and pyrimidine composition in 5S rRNA and its mutational significance Genet. Mol. Biol.

Variations observed in 5S rRNA base compositions are almost entirely due to fixation of point mutations. As a consequence, 5S rRNA size has remained relatively constant during evolution and, therefore, dependencies among the four bases can be predicted. In order to characterize the nature and to determine the degree of such dependencies, correlation analysis followed by principal component factorial analysis was conducted on a large sample of 5S rRNA sequences. The results show that the purine and pyrimidine contents tend to remain constant, so that A + G = Kpur and C + U = Kpyr. Interspecific hybridization and inbreeding effect in seed from a Eucalyptus grandis x E. urophylla clonal orchard in Brazil Genet. Mol. Biol.

We used allozyme markers to estimate the amount of natural hybridization between Eucalyptus grandis and E. urophylla in a 7.4-hectare commercial hybrid-seed orchard planted in Espírito Santo, Brazil. This orchard was planted in 1982 using a honeycomb design, with each hexagonal plot containing one E. grandis tree surrounded by six E. urophylla trees. There were 267 replicated hexagonal plots in the orchard. Seeds were harvested from the E. grandis clone only. The multilocus outcrossing rate estimated for the E. grandis clone averaged 70.2%, ranging from 33.0 to 99.0% among individual trees. Contaminant pollination, inferred from progeny genotypes containing alleles not present in the seven parental clones, accounted for 14.4% of the hybrid seed....

Plasminogen activator inhibitor-1 (PAI-1) is an important inhibitor of fibrinolysis, and increased levels of PAI-1 are associated with atheroma and myocardial infarction. A common 4G/5G insertion/deletion polymorphism located in the promoter region of PAI-1 gene has been described associated with PAI-1 activity in plasma levels. Genotyping of this polymorphism is commonly conducted with an allele-specific oligonucleotide melting technique. In the present study, we describe a quick, easy method for genotyping 4G/5G polymorphism in the promoter region of the PAI-1 gene.

Seventeen patients thought to have lethal osteochondrodysplasias were evaluated. Diagnosis was established through clinical evaluation, radiological studies and necropsy. Genetic counseling was provided to the affected patient's families. Specific diagnosis was confirmed in 16 cases. Nosologic diagnosis was done through clinical evaluation. However, the most efficient method for verifying the diagnosis was a skeletal radiological study. This fact corroborates the orientation of the International Classification of Osteochondrodysplasias (International Working Group on Constitutional Disease of Bone, 1992) in which a radiological criterion was adopted as the most relevant for classification of osteochondrodysplasias. Metaphase karyotypes of Anopheles (Nyssorhynchus) darlingi and Anopheles (N.) nuneztovari from Manaus and Highway BR-174 (Manaus-Boa Vista), State of Amazonas, and Macapá, State of Amapá, Brazil, showed 2n = 6. They consisted of a pair of metacentric (chromosome II) and a pair of submetacentric (chromosome III) autosomes as well as sex chromosomes X and Y. In the sex chromosomes, the X was acrocentric in A. darlingi and submetacentric in A. nuneztovari. The Y chromosome was pointed in both species. An important trait for common bean (Phaseolus vulgaris L.) cultivars with Carioca type grain is resistance to Colletotrichum lindemuthianum, which causes anthracnose and a pale corona. The present study was conducted to understand the genetic control of common bean reaction to the fungus and of the corona color, to provide guides to future breeding studies. Genotypes P-45, with brown corona, and EMGOPA 201-Ouro, with yellow corona, are resistant to C. lindemuthianum. Grain proteins from 58 Brazilian and nine Japanese upland rice cultivars (Oryza sativa L.) were electrophoretically separated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). Densitometric scanning of the electrophoretic profiles permitted the estimation of the relative concentration of 16 protein fractions, which were used as variables for the calculation of Fisher's canonical discriminating functions. Strain intercross experiments provide a powerful means for mapping genes affecting complex quantitative traits. We report on the genetic variability of the intercross of the Large (LG/J) and Small (SM/J) inbred mouse strains as a guide to gene mapping studies. Ten SM/J males were crossed to 10 LG/J females, after which animals were randomly mated to produce F1, F2, and F3 intercross generations. In the present paper we show data obtained from a normal population with a racially mixed profile typical of the city of São Paulo, State of São Paulo. Data were generated with polymerase chain reaction using sequence specific primers (PCR-SSP) for HLA-DRB and polymerase chain reaction followed by hybridization with sequence specific oligonucleotide probes (PCR-SSO) for HLA-DQA1 and HLA-DQB1 loci.
